Quantitative Proteomics Analysis of FFPE Tumor Samples Reveals the Influences of NET-1 siRNA Nanoparticles and Sonodynamic Therapy on Tetraspanin Protein Involved in HCC

Hepatocellular carcinoma (HCC) poses a severe threat to human health. The NET-1 protein has been proved be strongly associated with HCC proliferation and metastasis in our previous study. Here, we established validated the siRNA nanoparticles system conduct targeted gene therapy of xenograft vivo aid sonodynamic therapy. Then, conducted label-free proteome mass spectrometry workflow analyze formalin-fixed paraffin-embedded samples collected this result showed that 78 proteins were differentially expressed after inhibited. Among them, expression 17 upregulated 61 significantly downregulated. Of abundance, vast majority Gene Ontology enrichment terms belong biological process. KEGG pathway analysis enriched 45 pathways. We concluded function is not only regulate but also participate variety biochemical metabolic pathways body. Furthermore, protein–protein interaction indicated interactions are incredibly sophisticated. All happened silenced. Finally, study provides useful proposal for based on tetraspanin treat HCC, further mechanism investigations needed reveal more detailed action regulation HCC.
